How MOTS-c links mitochondrial genetics, AMPK signaling, and preclinical metabolic research.

MOTS-c is a 16-amino-acid mitochondrial-derived peptide reported from a short open reading frame within the mitochondrial 12S rRNA region. It is studied as a signal connecting mitochondrial state with nuclear and cellular metabolism.

What researchers examine

The discovery paper used cultured cells and mouse models to study folate-cycle and purine-biosynthesis changes, AMPK activation, insulin sensitivity, and metabolic homeostasis. Later work has examined nuclear translocation and stress-responsive gene expression.

How to interpret the evidence

The strongest mechanistic evidence is preclinical. Results in engineered cells and mouse models do not establish a human benefit, safety profile, or appropriate exposure. MOTS-c is also unusually sensitive to sequence and analytical identity because a short peptide can be altered substantially by a single residue, terminal modification, oxidation, or impurity.
